4 Sodium retention in microgravity (SOLO) PI: Martina Heer, PhD CoI: DLR: L. Beck, P. Frings, N. Baecker, Panum Inst., Copenhagen: P. Norsk, MD NASA JSC, Houston: S.M. Smith

A. Maillet, POIWG#25, Feb 3-5, Stimulus: - 2 different levels of salt intake (low, average normal) for 5 days each - randomized regarding the sequence of salt intake Schedule: - prefered: later in the mission - study phases close together Blood samples: day 5 24 h Urine sample: day 6 (final void: day 6 morning void) Body mass: day 4 and 6 SOLO STUDY DESIGN

A. Maillet, POIWG#25, Feb 3-5, In-flight measurements SOLO 6 days > 4 weeks >1month Session 1 Crew Member Launches 1< days <7 Session 2 Crew Member Lands Day 2Day 5Day 4Day 3 Day 1 Session Breakdown (1 or 2) Body Mass Measurements with SLAMMD – POIC Planning Blood Sampling with RC: blood draw and blood stow - ESA Planning; RC processing and MELFI insertion – POIC Planning Day 6 Controlled Diet Monitoring (random low or normal salt intake) – ESA Planning Diet and Exercise Log – ESA Planning Blood Sampling with PCBA: blood stick and PCBA measurement - ESA Planning Urine Sampling: 24-hr urine collection - ESA Planning; MELFI insertion – POIC Planning MELFI PCBA Retrieval – POIC Planning PCBA Software Update – ESA Planning This new activity requires to have the EPM ON, It is planned to link this with other experiments which require also EPM (for example DOSIS) NEW ACTIVITY vs. INCR-17:

A. Maillet, POIWG#25, Feb 3-5, SOLO HARDWARE Already on-board EPM CDL PCBA Kit (EPM Cardiolab Portable Clinical Blood Analyzer) Required for each increment: Need for PCBA SW update  File uplink via EPM  Use of EPM and Cardiolab GCP

A. Maillet, POIWG#25, Feb 3-5, SOLO HARDWARE Upload on 34P PCBA Consumables‘ Kit including the PCBA Cartridges Bag (cold 4°C) Solo Blood Sample Kit Solo Urine Collection Kit Already on-board EPM CDL PCBA Kit (EPM Cardiolab Portable Clinical Blood Analyzer)

A. Maillet, POIWG#25, Feb 3-5, Long-term microgravity: A model for investigating mechanisms of heart disease and sympatho adrenal activity during long duration space flight CARD

A. Maillet, POIWG#25, Feb 3-5, Major changes from last CARD session: -24 hrs Urine Collection using ESA Urine Collection Kit. -Only one 24 hrs session pr. subject. -Holter BP recording session will have battery change and data storage after 1/3 part of session (i.e. end of day one) -Refrigerated centrifugation is not mandatory, but nice to have. Major changes

A. Maillet, POIWG#25, Feb 3-5, CARD Timeline Holter BP: Data Storage Battery Exch

A. Maillet, POIWG#25, Feb 3-5, CARD – HOLTER BP Activities As for SOLO, use of EPM and its Cardiolab Science Module Before CARD recording session starts:  Holter AA battery insertion  Setup of CARD Holter End of day 1:  Holter data transfer and storage  Holter AA battery exchange  Setup of CARD Holter On Day 2:  Holter data transfer and storage  Holter AA battery removal  End of session using CDL GCP

A. Maillet, POIWG#25, Feb 3-5, Integrated Carfdiovascular (ICV) OPERATIONS SUMMARY Inflight, resting echocardiograms (using the HRF Ultrasound) with ECG recording will be performed on flight days 14, 30, 135, and R-15 using real-time remote guidance. On FD75, an exercise echo session will be performed with echo measurements taken before and after exercise. Both the Resting and Exercise Echo sessions will be preceded or followed by 24 hours of ambulatory BP/Holter/Activity monitoring. For these sessions, subjects will apply electrodes and then don the HRF Holter Monitor 2, ESA Cardiopres, and two Actiwatches (one at the waist and one at the ankle). These devices will be worn for 24 hours after which the devices will be doffed and the data from all experiment hardware will be downloaded to the HRF PC and downlinked to the ground. In-flight exercise and medication logs will be obtained through data sharing.
