INTRODUCTION

 Nanotechnology is part of science and technology about manipulation of matter at atomic and molecular size so that 1 Nano = meter.  Properties of the same metal nanoparticles vary according to many factors such as size, charge, shape and stabilizing agent used.  Gold in its bulk form has long been considered an inert, nontoxic, bio-compatible, noble metal with some therapeutic (and even medicinal) properties.  When the size of the typical objects decreases into nanoscopic dimensions (nanometer dimensions), gold behaves very differently than in bulk and its safety.

 A basic principle of toxicology; if exposure occurs in sufficient quantities, all materials are toxic, i.e., many drugs that are beneficial at low doses are toxic at high doses.  Gold nanoparticles [AuNPs] have been widely used in current medical and biological research, including; targeted delivery of drugs, Imaging and diagnosis of many diseases, Intravenous contrast agent for imaging Photothermal therapy agents Noninvasive detection of lung cancer. And many other topics.

 Along this line, much experimental work has been done, which confirms the non-toxicity of gold NPs.  However, on the contrary, as much conflicting researches are also present, which revealed the toxicity of gold NPs.  The basic question to be addressed is: how toxic are gold nanoparticles at the potential concentrations at which they might be used for therapeutics?

MATERIALS AND METHODS

SSynthesis and Surface modification of Gold Nanoparticles: Colloidal gold nanoparticles were synthesized by the optimization of classical Citrate reduction method. Then, Surface modification of gold nanoparticles was necessary to increase its colloidal stability in physiological fluid by adding poly –ethylene glycol (PEG). CCharacterization of Gold Nanoparticles: The prepared gold nanoparticles were characterized by using: UV-Vis Spectrophotometer. Zetasizer. High Resolution Transmission Electron Microscopy (for determination of the size, shape and size distribution of the prepared nanoparticles).

 Thirty six- day old New Zealand white male rabbits, around 1 kg weight were divided into two groups;  Gp1 served as control and i.v injected with 1 ml saline.  Gp2 rabbits were i.v injected with single dose of 200 ppm of prepared gold nanoparticles.  On 3 rd, 5 th and 7 th day post inoculation five animals from each group were euthanized prior to which, three blood samples were collected.  Liver, kidneys, spleen and lungs were dissected out of the body, wiped off blood, weighted. Part of each organ was stored at -20°c till use. The other part was kept in buffered neutral formalin 10% for histopathological studies

 Hematological studies Full hemogram picture was evaluated for all animals including RBCs count, packed cell volume (PCV), hemoglobin concentration (Hb), total leukocyte count (TLC) and differential leukocytic count (DLC ).  Serum biochemical analyses Separated serum samples were used for determination of:  Total proteins  Albumin, Globulins and  Albumin/Globulin ratio.  Blood urea, creatinine levels.  The activities of ALT and AST as well as glucose concentration.

 Determination of gold distribution in tissue It was carried out in tissue homogenate prepared from frozen specimens of liver, kidneys, spleen and lungs using Inductively Coupled Plasma Mass Spectrometry ( ICP-MS ) Histopathological examination Formalin fixed tissue specimens of liver, kidneys, spleen and lungs were routinely prepared for H&E stained sections and examined microscopically.  Statistical analysis

RESULTS

Synthesis and Characterization of AuNPS. Tri sodium citrate was served the dual role of both: reducing agent to reduce the Au (III) ions to Au (0) then a stabilizer, by forming a layer of citrate ions over the AuNs surface. The characterization was carried out using:  UV-Visible Absorption Spectroscopy Spectrophotometer results of AuNPs characterization showing peak absorption (0.33) at wave length 518nm.

 Zeta potential: The zeta potential of citrated AuNPS was found to be around mV, verifying that gold nanoparticles were charged and stable.

GNPs administration to rabbits did not show any significant difference in all of the examined blood parameters compared to those of the Control group at different time intervals 3 rd, 5 th and 7 th day. GNPs administrated group showed mild significant increase in leukocytic count associated with significant mild absolute neutrophila at 3 rd day with significant monocytosis at 3 rd, 5 th and 7 th day post administration in comparison with control set. GNPs did not significantly influence the levels of total protein, albumin, globulin and A/G ratio when compare them with those of controls. Results of tissue distribution of GNPs The obtained data revealed that GNPs were accumulated mainly in liver and spleen and to less extent in kidney and lung.

Results of histopathological examinations:

In conclusion: The results of the present study revealed mild to moderate changes in the hematological, serum biochemical parameters as well as histological picture of the examined organs caused by the used size of GNPs. Despite these preliminary studies more accurate studies need to be taken for designing GNPs with less immunogenicity and cytotoxicity to be used in vivo.
