Procedure for making a wet mount

Procedure for making a wet mount Step One: Use the lens paper to clean a glass slide and coverslip. Step Two: Place the specimen that you wish to observe in the center of the slide. Step Three: Using a medicine dropper, place one drop of water on the specimen. Step Four: Position the coverslip so that it is at a 45 degree angle to the slide *Make sure that the water runs along the edge of the coverslip Step Five: Lower the coverslip slowly to avoid trapping air bubbles.

Procedure for making a wet mount cont. Step Six: If a stain solution will be added to a wet mount, place a drop of the staining solution to the microscope slide along one side of the coverslip. Place a small piece of paper towel on the opposite side of the coverslip. Step Seven: As the water evaporates from the slide, add another drop of water by placing the tip of the medicine dropper next to the edge of the coverslip, just as you would if adding stains or solutions to a wet mount. If you have added too much water, remove the excess by using the corner of a paper towel as a blotter. Do not lift the coverslip to add or remove water.
