The full-length human CYB561 mRNA, with its cognate 3′-UTR, was expressed under control of a eukaryotic cytomegalovirus promoter in HEK-293T cells, with or without cotransfection of an miR-1294 mimic. CYB561 mRNA abundance was quantified by using quantitative real-time polymerase chain reaction (qRT-PCR), and normalized to glyceraldehyde-3-phosphate dehydrogenase mRNA. (B) MicroRNA effects in PC12 chromaffin cells: hsa-miR-1294 mimicry on human CYB561 mRNA expression with its cognate 3′-UTR. The experimental protocol proceeds as in A.
