Damec Research Aps POIWG July 2008 CARD Long-term microgravity: A model for investigating mechanisms of heart disease and sympatho adrenal activity during long duration space flight Thomas A. E. Andersen Damec Research Aps

Damec Research Aps POIWG July 2008 Astronauts in space exhibit low blood volume, pressure and cardiac output. A decrease in blood pressure and cardiac output induced by spaceflight, are caused by a deficit in salt and fluid intake. Studies of symapthoadrenal activity in blood samples (blood plasma and thrombocytes) will be combined with measurements of cardiac output, blood pressure and heart rate. Damec Research Aps POIWG July CARD Timeline

Damec Research Aps POIWG July 2008 Subject is 15A-FE2 – K. Wakata -Is been introduced in increment 18 though a CEF from ESA -JAXA ICB approval was obtained in June 08 -Due to the req. of minimum2 month in orbit the session will slide in to 19. Major changes from last CARD session: -24 hrs Urine Collection. -Only one 24 hrs session pr. subject. -Refrigerated centrifugation is not mandatory, but nice to have. Major changes 1

Damec Research Aps POIWG July 2008 CARD Hardware CARD PFS Consumables Kit CDL Holter Blood Pressure Device (HLTA) For 24 hours BP monitoring with measurements every hour. Pulmonary Function System (PFS) and Gas Delivery System (GDS) and RVU For a set of double rebreathing manoeuvres every 4 hours for 24 hrs. 3 Refrigerated Centrifuge Blood Sample Kit MELFI Urine collection 1x 24hrs, sampling per void

Damec Research Aps POIWG July 2008 Blood & Urine handling  Blood Draws One blood draw of 2 blood samples per measurement day before the last Rebreathing.  Urine sample One 24 hrs urine sample  Centrifugation Use of the HRF Refrigerated Centrifuge (4°C) for the blood samples:  1 sample at 500G for 15 min without break  1 sample at 1800G for 15 min with break  Freezing Frozen stowage of blood samples at < -20°C in MELFI and at -20°C in Cryogem 2

Damec Research Aps POIWG July 2008 CARD Experiment conditions The following conditions apply to the experiment: Food intake The subject should eat regular/equilibrated over the Reference and Measurement days. The subject should not be dehydraded. Exercise No exercise should be performed on the measurement days. Medication No medications should be taken on measurement days. EVA No EVA should be performed on the four Reference days, as it involves heavy exercise and irregular food intake. Circadian Rhythm The subject should have major chance in their Circadian Rhythm during the Reference and Measurement days.
