Sickle Cell Disease Archetype of the Single Gene Disease. Mutation in the gene encoding the βchain of hemoglobin with substitution of valine for glutamic acid at position 6 (βGlu 6 Val). HbS polymerizes in its deoxy conformation due to a hydrophobic interaction between HbS strands, with the formation of long fibers. Process is indispensable to disease pathophysiology, but is an insufficient determinant of phenotype.
