Most important type of protein? Enzymes! What do they do? Enzymes = speed up chemical reactions in the body* Hint: end in “a-s-e”*
Enzyme Uses Show animations: http://www.kscience.co.uk/anima tions/anim_2.htm
Catalase Reaction: H202 ____ + _____
Factors Affecting Reaction? What factors could cause the rate of the reaction to change? (Increase or decrease) Think about it: person eating a hamburger What would cause him to eat (break down the meat) at a faster rate or slower rate? Now what about the enzyme catalase?
Possible Experiments Test the affect of… -Temperature & pH -Substrate concentration (amount of hydrogen peroxide) -Enzyme concentration (amount of yeast) …on the rate of enzyme activity
Journal setup Copy this format onto one page in your journal Factors Affecting Enzyme Activity Copy this format onto one page in your journal Enzyme Concentration Substrate Concentration Temperature pH
Demo 1: Substrate Concentration How will the amount of substrate impact the rate of activity? (Rate of production) Thoughts? Test: Change amount of hydrogen peroxide, keep enzyme amount the same Use site: http://www.mhhe.com/biosci/genbio/virtual_labs/BL_11/BL_11.html
Substrate Concentration* Describe! Substrate Concentration* As the substrate concentration increases, the reaction rate increases
Demo 2: pH How will the pH (acidity) impact the rate of activity? Thoughts? Test: Change the pH (acidity), keep hydrogen peroxide amt the same, keep the enzyme amt the same Use site: http://www.mhhe.com/biosci/genbio/virtual_labs/BL_11/BL_11.html
The reaction rate increases until optimal pH and decreases past optimal pH Just like temperature, enzymes have an ideal pH where they work best (ie the top of the graph)
Demo 1: Enzyme Concentration How will the amount of enzyme impact the rate of activity? (Rate of production) Thoughts? Test: Change enzyme amount, keep hydrogen peroxide amt the same Website model: http://www.kscience.co.uk/animations/model.swf
Enzyme Concentration* We know from our experiment that as enzyme = the reaction rate More enzyme = more chances to collide with substrate But eventually, what happens? Reaction levels off, because not all enzymes can find substrate! As the enzyme concentration increases, the reaction rate increases
Demo 4: Temperature How will the temperature impact the rate of activity? Thoughts? Test: Change the temperature of enzyme. Keep same amount of hydrogen peroxide, keep same amount of enzyme Website model: http://www.kscience.co.uk/animations/model.swf
The reaction rate increases until optimal temperature, and decreases quickly past optimal temperature Temperature* What did the results from the experiment show, when testing cold, room temp, and heated yeast? Enzymes have an ideal temperature range, because they are proteins When proteins get too hot, they denature (3D structure destroyed = unable to function), and the reaction stops
Enzyme Limits Our results showed that as enzyme concentration increased, so did the rate of reaction Does that trend go on indefinitely? If we added TONS of enzyme, would the reaction just happen faster and faster and FASTER???
What are the names of ORGANISMS that live in thermal hot springs?? Different Ranges* Extremophiles! Different enzymes have different ideal temperature ranges
Why go faster?
Enzyme Review* Type of biomolecule? Naming? Protein Ends in “ase” Function? Substrate interaction? Speed up reactions Lock & Key How? Temp/pH impact? Decrease activation energy Ideal conditions only (too hot = denature = don’t work)
Do enzymes stop working after one use? Let’s see… No! Enzymes are reusable, meaning they do not get used up during a reaction*
