Figure 1 (A) Insomnia patients have a smaller gray matter (GM) volume than control subjects in areas in the left orbitofrontal cortex (OFC) and precuneus. Red areas indicate clusters of at least 37 voxels with p values smaller than .001 in Statistical Parametric Mapping (SPM) analyses. Coronal (y = 26, left panel) and axial (z = −14, middle panel) slices show the affected orbitofrontal area, superimposed on the Montreal Neurological Institute (MNI)152 standard brain. The sagittal slice (x = 2, right panel) shows areas of the precuneus in the parietal lobe. For coordinates of the voxel with maximum Z value within each cluster, see Supplement 1. Similar clusters were observed with nonparametric SnPM analyses (Supplement 1). (B) Regression analysis within the group of insomnia patients shows a strong negative relation between GM volume in the OFC and severity of insomnia measured by the Sleep Disorders Questionnaire (SDQ)-insomnia subscale. Green voxels indicate the OFC area where GM volume correlated significantly with severity of insomnia, with the strongest regression coefficient of b = −.71 at voxel x = −20, y = 22, z = −14. The green area overlaps with the OFC area where main effects analysis indicated a smaller GM volume in patients compared to control subjects (shown in red voxels). (C) Severity of insomnia is negatively related to left orbitofrontal GM density.
