48” 36” Background Objectives Methods In vitro Protein Tyrosine Phosphatase 1B Inhibitory Effect of Selected Medicinal Plants Y. Saidu, S. A. Muhammad*, L.S. Bilbis, B. S. Muhammad and L. Muhammad Department of Biochemistry, Usmanu Danfodiyo University, Sokoto Background Results Fractionation of the extract of S. nigricans partitioned the active agent significantly in the methanol fraction with 56.43±2.30% inhibition (Figure 2). This was comparably better than the value obtained for a known inhibitor of PTP 1 B, sumarin Protein tyrosine phosphatase 1B (PTP 1B), which is important in down regulation of insulin receptor has emerged as one of the best-validated drug target for type 2 diabetic management1 Natural products which contain inherently vast structural diversity are potential sources of lead compounds for the development of new therapeutic strategies for PTP IB inhibition2. The results (Figure 1) indicated that S. nigricans and Azadirachta indica had the highest percentage inhibition of 68.2% and 67.4% respectively. Objectives 36” To Screen for PTP IB inhibitors from medicinal plants used for management of diabetes mellitus Methods Figure 2: % Inhibition of Crude Fractions of S. nigricans Figure 1: % Inhibition of Crude Methanol Extract of Different Medicinal Plants Conclusion Methanol extracts of Mangifera indica, Moringa oleifera, Acacia nilotica, Arachis hypogea, Senna nigricans and Azadirachta indica screened for PTP 1B inhibition using PTP 1 B drug discovery kits, a product of Enzo Life Sciences. The methanol extract of S nigricans was further fractionated using separation funnel successively with hexane, ethyl acetate and methanol. The results were compared with a known inhibitor, sumarin The work demonstrated the potential of S. nigricans as a source of lead compound(s) in the development of inhibitors(s) of PTP 1B in the management of type 2 diabetes mellitus ( ( References Tran, H.Q.
