Directions 1. Delete the gel picture on the third slide. 2. Paste your gel picture on the third slide, crop to get your side of the gel, & resize so that the picture is about the size of the example on the third slide. The picture must be in black and white. 3. Re-align the standard lines on the left. Remember the biggest gap is between 1000 and 1650. You may need to re-adjust the lanes as well. 4. Replace the “#” in the top left with the proper clone round. Label the lanes with the proper clone name initials (i.e. change XX to student initials like BP or ZK). 5. Indicate the calculated size of the insert at the bottom of the gel (i.e. change ### to 500 bp). Note: remember to subtract the vector length of 200 bp from where the unknowns match up to the standard.

Directions ***If you cannot determine the size of your insert, put either contam (if the prep is contaminated) or N/A (if there are other problems). Make sure to explain why you can’t determine the size in a comment on the Clone Report Sheet. Refer to the PCRG Mockup Recap on the website if you are having trouble determining the problem*** 6. Complete all other fields like student names, date, and lab group #. 7. Save file as a .jpeg. Upload to your personal Google drive account and name the file with the proper PCRG name (i.e. 04PCRG1.14 Group 0). 8. Change the privacy settings of the file to “Anyone with the link can access” and paste the link on the Clone Report Sheet. Fill out approximated insert sizes on the Clone Report Sheet. If you cannot determine the size, explain why in a comment (right clickinsert comment).
