Trafficking-dependent and -independent modes of serotonin transporter regulation Depicted are 2 recently elaborated pathways that appear to establish appropriate levels of SERT transport activity in native models, neurons, and transfected cells. Trafficking-dependent and -independent modes of serotonin transporter regulation Depicted are 2 recently elaborated pathways that appear to establish appropriate levels of SERT transport activity in native models, neurons, and transfected cells. The trafficking pathway is dependent on protein kinase G (PKG) activation and can be triggered by receptors and mimicked by application of membrane-permeant PKG agonists such as 8-Br-cGMP. PKG activation also leads to a trafficking-independent mode of SERT regulation via p38 MAPK. Activation of p38 MAPK can also be triggered by anisomycin application and is followed by the conversion of surface SERT to a more active state. p38 MAPK inhibitors can block the activation process but do not impact the 8-Br-cGMP-triggered change in SERT surface density.
