Aim of the study To evaluate quality of Pre-Cut corneas for DSAEK prepared in the Interantional Eye Bank of Prague To analyse possible complications during preparation of the Pre-Cut corneas. To analyse possible complications after preparation of Pre-Cut corneas
Methods - preparation Pre-Cut corneas were prepared from 30 corneas – not suitable for surgery All Pre-Cut corneas were preapred in „Clean rooms“ by certified staff members All Pre-Cut corneas were prepared using standardized method for DSAEK – with Moria microkeratome Moria microkeratome Ljfpqejqoj tpoj Jkpavjpjp ojp
No major technical problems Epithelial abrasion Mounting Pachymetry No major technical problems during preparation Cut Free flap evaluatin Free flap reposition Dismounting
Methods - Evaluation All Pre-Cut corneas were evaluated using Slit lamp Specular microscopy Selected Pre-Cut corneas were screend for Microbial comtamination Ljfpqejqoj tpoj Jkpavjpjp ojp
Results - Slit lamp evaluation All 30 Pre-Cut corneas were evaluated using slit lamp In 28 Pre-Cut corneas we found good position of the corneal free flap In 2 cases we foud dislocation of the free flap into the medium No obvious stromal or endotehelial defects or opacity were detected Incorrect position of the free flap Ljfpqejqoj tpoj Jkpavjpjp ojp Correct position of the free flap
Results Specular microscopy evaluation The average endothelial cell censity prior PreCut was 2709±483(SD) cells/mm2, The average endothelial cell censity after PreCut was 2623±407(SD) cells/mm2 We found no significant endothelial cell loss after preparation. Corneal endothelium after Pre-Cut
Endothelial Cell Density Ljfpqejqoj tpoj Jkpavjpjp ojp
Resuts - Microbiology 5 randomly selected samples of medium and corneal swaps were microbiologically tested for microbial and fungal contamination – all results were negative no medium showed signs of bacterial contamination 1 week after preparation of the Pre-Cuts.
Summary Preparation Evaluation In our serie we experienced no major technical problems The procedure is straight forward with relatively short learning curve Evaluation In our serie evaluation of prepared tissue showed high quality and high safety
Conclusion We believe that our eye bank prepares Pre-Cut corneas for lamelar keratoplasty of very high quality and safety We also believe that in the near future Pre-Cut corneas will represent majority of tissues provided by our eye bank
