Lab 2: Preparing to clone the RFP gene: digesting the pKAN-R and pARA Title goes at the top of the first page Each section must be clearly underlined Skip lines between each section Use blue or black PEN! Follow these directions. You must be ready for lab (including pre-lab questions) at the beginning of the period. 1. Title: copy this title..." Preparing to clone the RFP gene: digesting the pKAN-R and pARA ” 2. Background: write a definition for the following words: plasmids, gene expression, selectable markers, pARA, pKAN-R, RNA polymerase, beta lactamase, ampicillin, fluorescent protein, restriction site. 3. Pre-lab questions: Answer the STOP and THINK question on page B-13. 4. Objective: Read the background information and determine the objective of the lab. 5. Hypothesis: Create your hypothesis. 6. Materials: copy the materials in a bulleted list 7. Procedure: Create a flowchart of the procedure. 8. Data : Take a picture of your tubes. Print and label. 9. Data Analysis: Copy and answer the STOP & THINK questions on page B-15 & B-16. 10. Conclusion: Copy & answer the 5 questions page B-17. (Do this after lab is over).
