268nm UV spectrophotometry for the Theorem Proving strings 2004.08.17 MEC SEMINAR Joon Shik Kim

*. Introduction. We measured the 268nm UV absorption in DNA strings for the Theorem Proving. -First measured the optical density as the temperature increases from 20 ’C to 90 ‘C. -Then measured the absorption of UV as decreasing the temperature from 90 ‘C to 20 ‘C -Temperature was controlled by water bath and spectrophotometer was BECKMAN COULTER(USA) DU 800 spectrophotometer. -NaCl 1M. -ssDNA 125pM, 250pM, 375pM.

**. Discussion. When the temperature decreases(backward), UV absorption at 80 ’C was larger than that of 90 ‘C. When the melting curves are compared on normalized form, forward curves of three density shows no difference but backward curves shows the difference. Large ssDNA density shows rather more efficient minimization of energy than that of lower DNA density. 4. Comparison with the simulation result is expected.
