Metformin: Restraining Nucleocytoplasmic Shuttling to Fight Cancer and Aging  Jorge Iván Castillo-Quan, T. Keith Blackwell  Cell  Volume 167, Issue 7, Pages 1670-1671 (December 2016) DOI: 10.1016/j.cell.2016.11.058 Copyright © 2016 Terms and Conditions

Figure 1 Inhibition of mTORC1 by Metformin By disrupting mitochondrial energetics, metformin alters passive transport through the nuclear pore complex. This prevents RagC from transiting through the nucleus, and acquiring the GDP-bound active state necessary for mTORC1 activation. Inhibition of mTORC1 increases transcription of ACAD10, in part through SKN-1/Nrf2. ACAD10 inhibits growth and extends lifespan in C. elegans, and reduces human cancer cell viability.
