TGF-β is also an anti-inflammatory cytokine TGF-β1-null animals demonstrate massive autoimmune inflammation affecting multiple organs. Systemic administration of TGF-β  prevents the relapse of autoimmune encephalomyelitis, insulitis and SLE in murine model. Puromycin aminoglycosise nephropathy in rat model: anti-TGF-β antibody  worsen albuminuria and proteinuria.

Hypothesis Circulating latent TGF-β 가 renal inflammation 을 protection 한다. TGF-β1 transgenic mice 에서 renal inflammation 의 prevention은 Smad7(Inhibitory smad) 의 upregulation과 관계 있다. Smad7 의 upregulation 이 NF-κB activation 을 inhibition 한다. NF-κB는 inhibitor인 IκBα 와 binding에 의해서 inactivation 하는데 Smad7 에의한 IκBα 의 induction 이 TGF-β1의 anti-inflammatory effect 의 key mechanism 이다.
