Plant tissue culture

Definition of Plant tissue culture : Plant tissue culture is a collection of techniques used to maintain or grow plant cells, tissues or organs under sterile conditions on a nutrient culture medium of known composition.

Brief History of plant tissue culture  Schelden and schwann make toti potency theory in  Successful Leaf culture by Haberlandt in  White first time established successful root culture of tomato in  Skoog and Miller demonstrated the rule of axin and cytokinin on root and shoot formation in tobacco – tissue in  Edward cocking enzymatic isolation and culture of protoplast  Murashige and Skoog introduced the medium for tobacco culture in Gottlieb Haberlandt Born28 November 1854 Mosonmagyaróvár Died30 January 1945 (aged 90) Berlin NationalityAustria FieldsBotany

Gisela 5 (rootstock for sweet cheery) GF677 ( rootstock for almond) Pyro dwarf ( rootstock for pear) Rootstock production in plant tissue culture section of PBTL

Gisela 5 Its Shoot proliferation have good result in DKW media with 0.5 mg/L BAP and 0.1 mg/L IBA for the root production MS half concentration Macro elements and half concentration sucrose with 0.6 mg/L IBA is perfect.

DKW media(Dampf-Kraft-Wagen )

GF677 Its Shoot proliferation have good result in GNH media with 0.1 mg/L BAP for the root production MS half concentration Macro elements and half concentration sucrose with 0.6 mg/L IBA and 4 mg/L thiamin HCl is perfect.

GNH(Garoosi, Nezami and Haddad)

Pyro dwarf Its Shoot proliferation have good result in QL media with 0.8 mg/ BAP/L and 0.08 IBA/L for the root production MS half concentration Macro elements and half concentration sucrose without hormone

QL(Quoirin and Lepoivre)
