A B Figure S1 Frequency allele of interest gDNA mtDNA gDNA+mtDNA Ref. populations A B Frequency allele of interest RG7155 Fab light chain RG7155 Fab heavy chain CSF-1 (dimer) CSF-1R (monomer) 4LIQ CSF-1R (dimer) 4WRM Figure S1 Figure S1. Spatial distribution maps of the two SNPs of interest by public genomic data and structural modelling of one of these with respect to CSF-1R ligand and RG7155 binding. A) Publicly available data from the 1000 Genomes Phase 3 set (25) and literature (26) were used to create spatial distribution maps on the Eurasian and African continents for both SNPs of interest via the Kriging interpolation algorithm applied by the Surfer® 12 software (Golden software, LLC). Current sample site is depicted by an asterisk and experimentally determined frequencies are listed in parentheses and adjacent isofrequency lines are labeled in accordance. B) Structural modelling for the rs10079250 SNP on position 362 of the CSF-1R ectoprotein was carried out by applying an overlay with the two following structures represented as pipes and planks: one for the monomeric CSF-1R bound to the Fab fragment of RG7155 (4LIQ; 8) and the other by a recently published structure (4WRM; 32) of the CSF-1R dimer bound to its ligand CSF-1. 4LIQ is additionally shown in its transparent surface structure. Color code is depicted and the amino acid exchange H362R is shown as spheres in its exact positioning with a 1.5X atom sphere radius.
