The antiapoptotic effect of CX3C chemokine fractalkine (CX3CL1) is independent of CX3CR1 genotype. The antiapoptotic effect of CX3C chemokine fractalkine (CX3CL1) is independent of CX3CR1 genotype. Genomic DNA was isolated from a subgroup of donors used in Figure 1C and genotyped for the 2 nonsynonymous single-nucleotide polymorphisms (SNPs) V249I and T280M in CX3CR1 using a polymerase chain reaction–based allelic discrimination assay. Only the 3 most common haplotypes were identified, and the response to CX3CL1 was analyzed by genotype. For each donor in each experiment, the response to serum-starvation was normalized to 100% and the effect of CX3CL1 expressed as a fraction of this to minimize technical variation between experiments.
