VEGF levels are rapidly increased in burn and CABG patients. VEGF levels are rapidly increased in burn and CABG patients. Plasma VEGF levels in burn (A) and CABG (B) patients were determined by ELISA. There was a rapid elevation (6 to 12 hours) of VEGF levels after trauma, followed by a gradual decrease over time (n=5 for burn, n=4 for CABG). Results shown are mean±SD for plasma VEGF levels, in both burn and CABG patient samples. Muhammad Gill et al. Circ Res. 2001;88:167-174 Copyright © American Heart Association, Inc. Expression of VEGFR2 and VE-cadherin mRNA in mobilized PBMNCs by RT-PCR. Expression of VEGFR2 and VE-cadherin mRNA in mobilized PBMNCs by RT-PCR. Representative sample from a burn (A) and CABG (B) patient, showing VEGFR2 (KDR) and VE-cadherin mRNA expression as determined by RT-PCR analysis of the PBMNCs. Muhammad Gill et al. Quantification of the mobilized VEGFR2+ and VE-cadherin+ cells by flow cytometry. Quantification of the mobilized VEGFR2+ and VE-cadherin+ cells by flow cytometry. Representative two-color flow cytometry analysis of mobilized PBMNCs, quantifying the number of VEGFR2+ and VE-cadherin+ cells 12 hours after burn injury (A) and 6 hours after CABG (B), demonstrating the number of VEGFR2+ (5.3% in burn, 11.1% in CABG) and VE-cadherin+ (4.7% in burn, 8.7% in CABG) cells. Time course of the total number of mobilized VEGFR2+CD15− cells in the PBMNCs of burn and CABG patients. Time course of the total number of mobilized VEGFR2+CD15− cells in the PBMNCs of burn and CABG patients. The total number of mobilized VEGFR2+CD15− cells over time in burn (n=8) (A) and CABG (n=7) (B) patients was determined by two-color flow cytometry in 1 mL of peripheral blood. Nonviable cells were excluded by PI staining. Muhammad Gill et al. Circ Res. 2001;88:167-174 Copyright © American Heart Association, Inc. All rights reserved.

Early and late endothelial outgrowth colony formation by the circulating mononuclear cells from burn (A) and CABG (B) patients. Early and late endothelial outgrowth colony formation by the circulating mononuclear cells from burn (A) and CABG (B) patients. PBMNCs (105 cells) obtained from the peripheral blood were cultured on gelatin-coated plates in the presence of endothelial growth medium. Adherent proliferating colonies that formed in the first 2 weeks and late colonies that formed in the passaged cells during 6 to 8 weeks of initial culture were quantified for the metabolic incorporation of DiI-Ac-LDL (red) as well as vWF (green) staining (C). DiI-Ac-LDL+vWF+-adherent colonies that formed from passaged nonadherent cells, between 6 to 8 weeks of initial culture, were scored as late-outgrowth colonies (CFU-ECs), whereas those that formed from initial plating, within the first 3 weeks, were considered as early outgrowth colonies. Acute VEGF elevation promotes rapid but transient mobilization of VEGFR2+ cells. Acute VEGF elevation promotes rapid but transient mobilization of VEGFR2+ cells. A, Two-color flow cytometry showing the number of VEGFR2+CD11b− cells mobilized 24 hours after AdNull or AdVEGF165 intravenous injections, demonstrating that 1.7% and 14.7% of VEGFR2+CD11b− cells were mobilized, respectively. B, Time course of the mobilized VEGFR2+CD11b− cells in the AdNull- and AdVEGF165-injected mice (n=3). The number of VEGFR2+CD11b− cells was determined by flow cytometry. Nonviable cells were excluded by PI staining. Muhammad Gill et al. Circ Res. 2001;88:167-174 Copyright © American Heart Association, Inc. All rights reserved.
