A, C2C12 cell lines which stably harbor cumate-inducible FLAG-WT-FoxO1 or FLAG-DN-FoxO1 were established as described in the Methods section in the Data Supplement. Cumate (300 μg/mL) was added to the culture medium for 48 hours to induce gene expression. Wnt3a (50 ng/mL), Dkk1 (200 ng/mL), or 0.1% BSA/PBS (control) was added to the culture medium for the last 24 hours. Total protein samples extracted from C2C12 cells were immunoprecipitated with an antibody against FLAG or control IgG and analyzed by Western blotting.
