When time does not heal wounds: Optical imaging of diabetic wounds PhD student: Shima Mehrvar Advisor: Dr. Mahsa Ranji Collaborators: Drs. Sandeep Gopalakrishnan and Janis Eells, Colleges of Nursing and Health Sciences, UWM 04/21/2018

Motivation Chronic wounds: Diabetic wounds: Deed for Cure: Nonhealing wounds More than 6.7 million people in US Diabetic wounds: Diabetic foot ulcers 15% of diabetic patients Deed for Cure: Diagnostic tools Wound care therapies Diabetic wound (mouse) First of all I would like to tell you our objectives for this project, which was to quantitatively evaluate the redox state of rat liver under ischemic and reperfusion conditions for transplant application This project caught our interest because “liver organs qualified for liver transplantation are in shortage and Evaluation of a liver for transplant is critical for success!” As you know before doing a liver transplant you need to figure out if the liver is a match to the patient. There’s several ways to do that now already, but sometimes there’s still problems and the liver gets rejected. In this project, we’re trying to use optical imaging technology to evaluate the metabolic state of a liver sample, both before, during and after the transplant to try to improve the current evaluations methods. With our techniques we can show 3D reconstruction of different regions which can quantify the metabolic health.

Real-time Optical Imaging In vivo optical imager Metabolic images Healing Mercury arc lamp CCD camera Day0 Day1 Day2 Day3 Day4 Filter wheel Diabetic Control Filter wheel The first method is fluorescence spectroscopy, which we call the instrument fluorometer. Fluorometer has the advantage that it can be used in vivo or ex vivo. It is nondestructive and portable. It can measure fluorescent intensities online and quantify the metabolic state of the tissue where the probe is closely against the beating heart. You can see schematic of the instrument, where the filtered light excite through a bifurcated fiber cable and emission signals were filtered and recorded by photomultipliers. The software will calculate the redox ration online on the screen Not Healing Noninvasive Portable system Online information Constant lower metabolic state (blue color) in diabetic wounds

Conclusion In vivo optical imager: Future direction: Online information about the tissue metabolism Noninvasive (visible light) Cost-effective Portable Future direction: Treatment options such as near-infrared light therapy
