DNA Extraction: Your choice of fruit

Does a(n) __________ have DNA? Question Does a(n) __________ have DNA? (pick a fruit that we did NOT do in class – no strawberries or kiwis)

Materials Ziploc bag 1 fruit of your choice 2 Tbs DNA extraction buffer (soapy, salty water) next slide has recipe Coffee Filter paper or paper towel Funnel Tall and skinny glass Chop stick or popsicle stick 3 Tbs ethanol or rubbing alcohol ICE COLD!

Create your DNA Extraction Buffer solution Mix: 1 cup water 1 teaspoon dish soap 1 teaspoon salt Be sure to put the rubbing alcohol in the freezer! It needs to be cold to do the experiment!

Procedure 1. Place 1 fruit in a Ziploc bag. 2. Smash up the fruit using your fingers for 2 minutes. Careful not to break the bag!! 3. Add the provided 2 Tablespoons of extraction bugger to the bag. 4. Mush the fruit again for 1 minute.

5. Assemble your filtration apparatus

Procedure 6. Pour the fruit slurry into the filtration apparatus and let it drip into your cup or test tube. 7. Slowly pour 3 Tablespoons cold ethanol or rubbing alcohol into the tube. OBSERVE 

8. Dip your stir stick into the tube where the fruit extract and ethanol layers come into contact with each other. OBSERVE! 

Throw away CLEAN UP! Wash and return Paper towels Strawberry goo Filter paper Stir sticks Funnel Test tube Graduated cylinder Any other lab equipment
