Supplementary Figure 1 Supplementary Figure 1. Dopamine depletion of the dorsolateral striatum. Sections were washed repeatedly in PBS and PBS containing 0.3% Triton X-100 (PBS/Triton), and then incubated in blocking buffer (5% normal goat serum, 0.2% cold white fish skin gelatin, PBS/Triton) for 1 hour. Sections were then incubated with mouse anti-tyrosine hydroxylase (1:1000; Millipore; Billerica, MA) for 48 hours at 4°C. They were subsequently washed five times in PBS/Triton before being incubated with biotinylated goat anti-mouse IgG (1:100; Vector Laboratories; Burlingame, CA) for 1hr. Sections were washed in PBS/Triton and developed with an avidin-biotin complex stain (VECTASTAIN ABC Elite Kit, Vector Laboratories) using a nickel-enhanced diaminobenzidine reaction. (a) Exemplar sections from a saline- and 6-OHDA-treated rat showing normal and reduced TH staining in the dorsolateral striatum, respectively. (b) Schematic illustration of coronal sections of the rat brain showing the minimum (dark grey) and maximum (light grey) regions of tyrosine hydroxylase (TH) depletion in the dorsolateral striatum of 6-OHDA-treated rats.
