Workflow of one-step integration into any potential chromosome location. Workflow of one-step integration into any potential chromosome location. (A) After NotI digestion, the camphor trans-activator is ready to be integrated into Chr XI between GAT1/YFL021W and PAU5/YFL020C. The camphor promoter can be easily amplified by PCR with the primer containing the 40 bp homologous region (green) from the target chromosome location for the integration and 20 bp region (magenta) for annealing. Based on the one-step integration strategy, the LYS2 gene was used to confirm the efficiency of shutoff. Cells were cultured for 1 d at 30° in SC–Lys (B), SC–Lys +100 µM camphor (C), and SC with 100 µM of camphor (D). The cells were diluted 10-fold in across each row. Shigehito Ikushima et al.
