Figure 1 FER Negatively Regulates Seed Size. (A)fer muants produce larger seeds than the wild-type. The fer mutants and wild-type control were grown under the same conditions and seeds were harvested at the same time. Photos of dry seeds were taken 1 week after harvest. (B)fer mutants show larger cotyledons than wild-type. Seeds were germinated on MS agar plate and photos were taken 4 d after germination. (C) Analyses of different cell types in the seeds. (a, b): cleared whole-mount seeds of wild-type and fer-4 imaged at globular embryo stage. Comparison of cell number (c), and average length of cells (d) in the abaxial oi2 and ii1’ layers of integuments in wild-type and fer-4 seeds. Bars = 20 μm. (D) Expression of pFER::FER–GFP in integuments. (a, f): ovules before fertilization, arrow indicates synergid. Globular embryo stage seeds (b, c, g, h) and a close-up image of FER–GFP in the abaxial integument cells (k, l, red arrows). Seeds at the torpedo stage (d, i) and cotyledon stage (e, j). Bars = 20 μm. (E) Seed size in GEF1-overexpressing plants (GEF1–OX). The GEF1–OX and wild-type control were grown under the same conditions and seeds were harvested at the same time. Images of dry seeds (upper) and cotyledons (lower) of GEF1–OX mutants and wild-type were taken under stereoscopic microscopes. (F) Average weight of GEF1–OX and control seeds. Each data point shows the average ± SE of four replicates per genotype containing 100 seeds. (G) Expression of ProGEF1–GUS in a seed at the globular embryo stage. Bar = 50 μm.
