Figure 1 Recognition of PfCSP Repeating Motifs by Human mAbs Top: Schematic representation of mAbs CIS43 and MGG4 binding to PfCSP. PfCSP contains an N-terminal domain (NTD), region I (RI), and a tandem repeat consisting of NPDP, NVDP, or NANP motifs followed by a C-terminal domain (CTD) and a GPI anchor. mAbs CIS43 and MGG4 demonstrate cross-reactivity with the NPDP, NVDP, NANP, and CTD domains, resulting in multiple mAbs capable of binding to a single PfCSP molecule. Fabs are shaded according to their binding affinity to the different repeating motifs (Kisalu et al., 2018; Tan et al., 2018): high-affinity binding is shaded dark, and lower-affinity binding is shaded light. Bottom: Comparison of PfCSP NPDP, NVDP, and NANP repeating motif recognition by mAbs CIS43 (Kisalu et al., 2018), MGG4 (Tan et al., 2018), 663 (Triller et al., 2017), 580-gl (Triller et al., 2017), 311 (Oyen et al., 2017), and 317 (Oyen et al., 2017). Common structural features, including Type I β turns (white circles), exist. However, all mAbs bind different peptide conformations, highlighting the existence of several immune solutions that engage the repeating motifs and inhibit Pf.
