Fig. 5 NF-κB heterodimer p50/p65 complex binding on DNA: inhibition with ASU. Bandshift assays were performed as described in Materials and methods. Mouse costal chondrocytes were cultured and grown to confluence and serum starved for 24h followed by 10μg/ml ASU incubation during 48h. Then, cells were incubated in the presence or absence of IL1β 10ng/ml for 0–15min. A 100-fold excess of unlabeled oligonucleotide consensus sequence wt over the NF-κB probe concentration was added to the nuclear proteins for 15min before adding the NF-κB probe. Antibodies (1μl of pure stock solution) raised against p50 or p65 were incubated with chondrocytes nuclear extracts for 15min at 4°C prior to adding poly (dI–dC). Nuclear extracts were then submitted to electrophoresis run on a non-denaturing 6% acrylamide gel in Tris–Borate.
