First, calculate amount of Chl a Ca = 11.85A665 - 1.54A647 - 0.08A630 Next, calculate mg Chl a / m3 mg Chl a/m3 = (Ca * v) (V * p) v = 10mL (acetone + SW) V= 1 mL (SW) p = 1.6 (path length)

Amount in 5 mL homogenate? We have mg / m3 0.01cm / m 106 cm3 / m3 cm3 = mL 106 mL / m3 Now we have mg / 106 mL Rearranging gives us 10-6 mg / mL Multiplying by 5 mL gives mg Chl a in the homogenate sample

We’re interested in the amount of Chl a in relation to the amount of zoox in the sample Divide the mg Chl a just calculated by the zoox concentration determined from the hemocytometer mg Chl a 106 zoox
