Note the bidirectional migration of progeny away from active stem cells in the isthmus (narrowing) of the gastric unit with longer lived cells in the base and the stem cell zone nearer the stomach lumen than the base. B, Matsuo et al find Runx1 promoter elements (eR1) drive expression of a reporter (blue) in the isthmus, with rarer cells in the base. Expression of constitutively active, oncogenic K-Ras by eR1 most commonly resulted in the overproduction of surface pit (foveolar) cells (lavender, top) likely driven by isthmal eR1. Rarer gastric units developed proliferative SPEM-type metaplasia likely driven by K-Ras from basal eR1-expressing chief cells.
