Figure 3 The effect of hormones on Jeg-3 spheroid–Ishikawa endometrial cell coculture assay. (A) The concentrations of E2 and P used in the coculture study are shown. (B) Jeg-3 spheroids of sizes 60–150 μm were selected and cocultured with (P4 or E2) or without (Neg) hormone-treated Ishikawa cells. Hormonal concentrations that resemble the serum levels of patients in natural cycles (N) or moderate responders of a stimulated cycle (M) do not affect spheroid attachment in vitro. However, hormonal concentrations that resemble those of excessive responders of a stimulated cycle (E) significantly down-regulate spheroid attachment onto Ishikawa cells in vitro.
