The oxidation of dihydrorhodamine 123 to rhodamine after stimulation with phorbol 12-myristate 13-acetate (PMA) is measured by an increase in fluorescence intensity. Granulocytes of the healthy control show an increase of fluorescence from 6 to 429, corresponding to a stimulation index of more than 40. The patient's granulocytes show a decreased stimulation index of 2, which indicated a severe reduction of ROS production.
