Parasite Clearance in Leishmaniasis in Resistant Animals Is Independent of the IL- 23/IL-17A Axis  Kirsten Dietze-Schwonberg, Beate Lorenz, Susanna Lopez Kostka, Ari Waisman, Esther von Stebut  Journal of Investigative Dermatology  Volume 136, Issue 9, Pages 1906-1908 (September 2016) DOI: 10.1016/j.jid.2016.05.111 Copyright © 2016 The Authors Terms and Conditions

Figure 1 IL-23p19 and IL-17A are irrelevant for L. major clearance in Leishmania-resistant C57BL/6 mice. (a, d) IL-17A−/−, IL-23p19−/− mice (both on a C57BL/6 background) and CD4-IL-17Aind/+ mice with artificial overexpression of IL-17A in CD4+ T cells were infected with 103 metacyclic promastigotes in both ears. Wild-type C57BL/6 or IL-17Aind/− mice served as controls. Lesion volumes were measured weekly in three dimensions. (b, e) Parasite burden in ears and spleen in weeks 6 and 9 after infection was assessed using limiting dilution assays. Dots/rectangles represent single organs, and bars indicate means. (c, f) Draining lymph node cells from infected IL-17A−/−, IL-23p19−/−, and C57BL/6 control mice, or CD4-IL-17Aind/+ and IL-17Aind/− control mice were restimulated with soluble Leishmania antigen (SLA) for 48 hours. Levels of IFN-γ, IL-4, and IL-10 in supernatants were assessed by ELISA. (a–f) Data are presented as mean ± standard error of the mean, n = 2–4 independent experiments with n ≥ 4 mice/group (*,+P ≤ 0.05, **,++P ≤ 0.005, ***,+++P ≤ 0.002 with * comparing against C57BL/6, + comparing CD4-IL-17Aind/+ mice vs. IL-17Aind/− mice).
