Axotomy results in the rapid degradation of NMNAT2, resulting in an increase in its substrate, nicotinamide adenine mononucleotide (NMN). SARM1, downstream (or in a parallel pathway) of NMNAT2 loss, is activated, which leads to axon degeneration. The axon-protective effects of NMNd may be due to lowering NMN levels by using it as a substrate for the synthesis of NaMN.
