Figure 1 Model for the protective effects of NMN deamidase (NMNd). Axotomy results in the rapid degradation of NMNAT2, resulting in an increase in its substrate, nicotinamide adenine mononucleotide (NMN). SARM1, downstream (or in a parallel pathway) of NMNAT2 loss, is activated, which leads to axon degeneration. The axon-protective effects of NMNd may be due to lowering NMN levels by using it as a substrate for the synthesis of NaMN.
