Volume 118, Issue 5, Pages 835-841 (May 2000) Microsatellite instability and mutation of mitochondrial and nuclear DNA in gastric carcinoma  Wataru Habano, Tamotsu Sugai, Shin–Ichi Nakamura, Noriyuki Uesugi, Toru Yoshida, Shunichi Sasou  Gastroenterology  Volume 118, Issue 5, Pages 835-841 (May 2000) DOI: 10.1016/S0016-5085(00)70169-7 Copyright © 2000 American Gastroenterological Association Terms and Conditions

Fig. 1 (A) mtMSI in the (C)n tract was detected by PCR-SSCP analysis. Cases 17, 23, and 28 show differing mobilities in carcinoma tissues (C) compared with normal counterparts (N). These alterations were detectable in both single-strand (ss) and double-strand (ds) DNA patterns. Case 28 was defined as mtMSI negative because the mutant sequence could not be determined by direct sequencing. (B) Mutations in the mitochondrial tRNA (Leu) and ATPase 6 genes were detected by PCR-SSCP analysis. Cases 5 and 23 showed mobility-shift bands (arrowheads) in comparison with normal counterparts, whereas others (4 and 24) did not show any band shifts. Fig. 2 Case 5 showed nMSI-H phenotype at the BAT26 and D2S123 markers and mutations in the TGFβRII (A)10 sequence. Arrowheads in carcinomas B, D, and F indicate mutant signals, which were not observed in normal mucosa (A, C, and E, respectively). Fig. 3 (A) mtMSI, (B) nMSI-H, and (C) p53 mutation or LOH were compared with the stage of invasion. m, intramucosa; sm, submucosa; mp, muscularis propria; ss<, subserosa or exposed at the serosa.
