FIGURE 1 Multiplex RT-PCR of tPA and β-actin in adhesion and peritoneal fibroblasts under normal and hypoxic conditions. (A), Multiplex RT-PCR of tPA and β-actin was performed as described in the methods section using total RNA isolated from normal peritoneal fibroblasts before (lane 1) and after hypoxia (lane 2) and adhesion fibroblasts before (lane 3) and after hypoxia (lane 4) treatment. Polymerase chain reaction products were analyzed on a 2% agarose gel and visualized by ethidium bromide staining. (B), The density of each band was measured by a scanning densimeter. Relative pixel densities corrected to internal control (β-actin) were used to compare the effect of hypoxia treatment on tPA mRNA levels. Error bars represent the standard error of the mean of triplicates. Results are representative of three independent experiments (P<.05). Gray, control; black, hypoxic. M = molecular weight marker.
