Figure 1 A Chk1-Mediated Zygotic Checkpoint Monitoring DNA Damage Arising from DNA Demethylation During zygotic reprogramming, DNA demethylation occurs in the paternal genome in G1. Xrcc1Δ zygotes accumulate DNA damage in G1, but not in G2, allowing them to enter mitosis normally. Scc1Δ zygotes accumulate DNA damage in both G1 and G2, leading to a G2 arrest before mitosis. The Scc1-15KR mutant is proficient for sister chromatid cohesion in S and G2 but defective for DNA repair in G1. Expression of Scc1-15KR in Scc1Δ zygotes suppresses the DNA damage arising from cohesion defects, but not repair defects in G1. Chk1 inhibitor (Chk1i) overrides the G2 arrest in Scc1Δ zygotes, showing that demethylation-associated DNA lesions trigger a Chk1-mediated checkpoint if they are not repaired.
