Figure 1 The Interaction between GATA4 and TBX5 in Cardiac Gene Regulation (A) WT cardiomyocyte transcriptional regulation is modulated by GATA4-TBX5 co-binding to MED1-bound cardiac super-enhancers activating key cardiac genes. Simultaneously, HDAC2 is recruited, resulting in a closed chromatin state at endothelial promoters. (B) The GATA4 G296S missense mutation suppresses the expression of cardiac specific genes by changing the proximity of TBX5 to GATA4. A failure to recruit HDAC2 with a euchromatin structure at endothelial promoters allows for mislocalization of TBX5 and expression of endothelial genes.
