Figure 1 A salient stimulus, such as the honking horn of an oncoming truck, is thought to elicit a burst of thalamic activity. In this issue of Neuron, Ding et al. show that such thalamic stimulation excites a burst-and-pause response in the cholinergic interneurons of the striatum. The resulting burst of acetylcholine (ACh) causes a brief decrease in cortico-striatal synaptic transmission to medium spiny neurons (MSNs) in both the direct and indirect pathways, via activation of presynaptic M2/M4 receptors. This is then followed by prolonged facilitation of transmission in the indirect, but not the direct, pathway, caused by activation of postsynaptic M1 receptors on MSNs in the indirect pathway.
