Figure 1 cRHIMS Drive IMD Pathway Activation through Functional Amyloid Formation DAP-type PGN can activate IMD signaling either at the plasma membrane by binding to PGRP-LC or from the cytoplasm by binding intracellular PGRP-LE. Kleino et al. (2017) show that this leads to cRHIM-mediated nucleation, which must then incorporate the cRHIM of Imd to form a fibril. This presents the DD of Imd to the downstream signaling components including FADD, which interacts through its DD. Pirk contains a cRHIM that can interact with the cRHIMs of Imd or PGRPs and thereby block amyloid formation preventing signaling.
