Figure 1 Ultra-trace Analytical Method and its Application in Analyzing GAs in Tiny Organs of A. thaliana. (A) The scheme for the newly developed ultra-trace analytical method: extraction of GAs, EDC labeling, and LC-ESI-MS/MS analysis. IS, internal standard. (B) Matured flower from primary inflorescence of wild-type A. thaliana (Col-0) (left) and dissected stamens from the flower (right). The plants were grown in a greenhouse at 24°C with a 16-h photoperiod and 52% humidity for 30 days. Matured flowers (approximately stage 13–15) from primary inflorescence were harvested and immediately frozen in liquid nitrogen. Stamens (including filaments and anthers) were carefully dissected from flowers under frozen conditions. (C) Identified and quantified GAs in the whole flowers and stamens of A. thaliana measured on LCMS-8050 (Shimadzu, Kyoto, Japan). Samples of one flower were obtained from the same inflorescence; samples of six stamens were dissected from three different flowers; samples of two stamens and one stamen were dissected from one flower. The value in each block is the content (data are presented as the mean ± SD, n = 3) as pmol/g fresh weight. nd, not detected (S/N < 3); nq, not quantified (3 < S/N < 10). (D) Representative multiple-reaction monitoring chromatographs of quantified GAs from six stamens of fresh A. thaliana flower and their corresponding internal standards. Transitions of each analyte are as follows: EDC-GA8 (520.30 → 404.15), EDC-dGA8 (522.30 → 406.15); EDC-GA1 (504.30 → 388.20), EDC-dGA1 (506.30 → 390.20); EDC-GA34 (504.30 → 388.20), EDC-dGA34 (506.30 → 390.20); EDC-GA51 (488.30 → 372.15), EDC-dGA51 (490.30 → 374.15); EDC-GA4 (488.30 → 372.25), EDC-dGA4 (490.30 → 374.25).
