Autoradiographic mapping and characterization of insulin-like growth factor-I receptor binding in human greater saphenous vein  Anton N. Sidawy, MD, Fares S. Hakim, MD, Richard F. Neville, MD, Louis Y. Korman, MD  Journal of Vascular Surgery  Volume 18, Issue 6, Pages 947-954 (December 1993) DOI: 10.1016/0741-5214(93)90549-2 Copyright © 1993 Society for Vascular Surgery and International Society for Cardiovascular Surgery, North American Chapter Terms and Conditions

Fig. 1 A, Hematoxylin-cosin-stained frozen section of human greater saphenous vein. B, Autoradiograph of adjacent section demonstrating total 125I-IGF-I grain density corresponding to vein wall. Optical density of binding measures 0.37 ± 0.04. C, Nonspecific binding for insulin. Excess of insulin was used to compete with 125I-IGF-I for its binding sites. Optical density measures 0.15 ± 0.01. D, Nonspecific binding for IGF-I. Excess of unlabeled IGF-I was used to compete with 125I-IGF-I for binding sites. Optical density measures 0.05 ± 0.007, which is not different from optical density of film measured at 0.04 ± 0.004. Note difference in nonspecific binding between C and D. Difference in optical density of C and D autoradiographs is statistically significant with p < 0.005 with paired t test. Fig. 2 Scatchard plot of 125I-IGF-I bound/free as function of bound radiolabeled peptide. Each data point represents mean ± SEM of results of veins studied. Calculated Kd = 1 ± 0.32 nmol/L and Bmax = 3.64 ± 0.47 pmol/L (0.46 ± 0.23 pmol/mg protein). Fig. 3 Binding inhibition curves of IGF-I and insulin. Each value is represented as mean ± SEM of results obtained from veins studied. Curves were plotted between 125I-IGF-I bound as percentage of maximal binding and concentration of unlabeled IGF-I and insulin used to compete with 125I-IGF-I binding. Calculated IC50 for IGF-I and insulin were 0.069 μmol/L and 6.9 μmol/L, respectively.
