Supplemental Figure 1A – I D G B C E F H I Supplemental Figure 1: Dose-dependent cytotoxicity of IPI-145 in primary CLL cells (n=9; 24 hrs). Primary CLL cells were cultured in suspension in presence of increasing concentration of IPI-145 and % viability was measured by Annexin/PI binding method.

Supplemental Figure 2A – I D B C E F G H I Supplemental Figure 2: Time-dependent cytotoxicity of IPI-145 in primary CLL cells (n=9). Primary CLL cells were cultured in suspension in presence of IPI-145 and % viability was measured by Annexin/PI binding method at 12, 24 or 48 hr. The % viabilities are normalized to relative viability of time-matched control samples (100%) to account for differences in spontaneous apoptosis in samples.

Supplemental Figure 3A and B Supplemental Figure 3: Cytotoxicity of IPI-145 was tested in primary CLL cells at (A) as low as 500 nM (n=11) and (B) as early as 12 hrs (n=4). The % viability was measured using Annexin/PI binding assay.
