Figure 1 An Allosteric Structural Transition in S. pombe Poz1 and Human TIN2 Telomeric Proteins Drives the Formation of the Shelterin Complex Fission yeast and human shelterin subcomplexes that are either favored (upper panel) or disfavored (lower panel) by cooperative binding of subunits. For the sake of comparison with the human shelterin, the Taz1-Rap1 subcomplex is depicted as a single building block. The allosteric transition in Poz1 (TIN2) “conformational trigger,” induced by Tpz1 (TPP1) binding, increases Poz1 affinity for Rap1-Taz1 (TRF2), making the assembled protein bridge a more stable entity than individual subcomplexes (upper panel) and decreasing the probability for the existence of the subcomplexes depicted in the lower panels.
