Figure 1 Principal component analysis (PCA) representing all the metabolites analyzed for each sample in the two rounds. Blue/green triangles represent fresh group, and red triangles represent vitrification group. No separation between groups is observed using (A, C) positive and (B, D) negative ionization of the samples analyzed in both rounds of analysis. Figure 3 (A) Box plots of alpha-CEHC, a metabolite found to be statistically significant between fresh and vitrification groups using positive and negative ionization. Dots correspond to individual measurements of alpha-CEHC. (B) Receiver operating characteristic (ROC) curves are shown for this metabolite. Supplemental Figure 1 Experimental design of the study. Microdrops were taken from 190 spent culture media from embryos that had been cultured from day 1 to day 3. Collection was performed on day 3 by collecting 50 μL of culture media. The samples were collected from the fresh group (n = 59), the vitrification group (n = 65), and matched control media group (n = 66) in the LC-MS study. Metabolomic analysis was performed in two rounds due to the instrument capacity limitations.
