Fig. 1 Ileal biopsy specimens from patients with Imerslund–Gräsbeck syndrome and ileal mucosa obtained from an unidentified donor were homogenized in Tris-HCl buffer (pH 7.4) containing 140 mmol/L NaCl and centrifuged at 100,000g for 1 hour. The resulting pellet was resuspended in the same buffer and homogenized. Total membrane protein (50 μg) containing 2 fmol (lane 1, control), 25 fmol (lane 2, patient 1), or 5 fmol (lane 3, patient 2), or 100 μg protein containing 50 fmol (lane 4, patient 1), 10 fmol (lane 5, patient 2), or 20 μg of total membranes from 14-day-old pregnant rat yolk sac containing 20 fmol of ligand binding activities (lane 6) were subjected to 5% sodium dodecyl sulfate–polyacrylamide gel electrophoresis.
