Figure 2 Morphogenesis of Y224 (A-C) and M634 (D-H) cells cultured in 0.1% type I collagen gels. (A) Y224 cells formed mostly cell aggregates within eight days. (B and C) Cross-section view of Y224 cell aggregates under hematoxylin and eosin staining examined by light microscopy. (D) M634 cells developed pseudopods in 0.1% collagen gel within 1 day. (E and F) M634 cells formed branching tubule structures within three and eight days, respectively. (G) A long tubule with lumen taken from an eight-day culture of M634 cells in collagen gel. (H) Cross-section of the M634 tubule formed within eight days in collagen gel. Panels A, D, E, F, and G were taken under phase-contrast microscopy. Scale bars, 100 μm (F) and 50 μm (A-E and G-H). Figure 3 Comparison of morphology (A-D) and migration capacity (E) between M634 and Y224 cells. M634 cells (A and C) and Y224 cells (B and D) were cultured on plastic plates (A and B) or collagen gels (C and D), and photographs of their colony morphology were taken under phase-contrast microscopy. Y224 cells formed connected cell islands with greater cell-cell association, whereas M634 cells exhibited higher degrees of scattering. (E) Collagen-induced cell migration capacity of M634 and Y224 cells. Migration assays were performed as described in the Methods section. There is a significant difference in the mean number of migrated cells per high-power field between M634 and Y224 cells (scale bar, 100 μm). Figure 4 Comparison of proteins in focal contact components between M634 and Y224 cells. (A) Detection of focal contact components in the total cell extracts was made by Western blot analysis. Cell extracts were prepared from cells cultured in 0.3, 0.2, or 0.1% collagen gels for eight days, and 30 μg protein was resolved by 7.5% SDS-PAGE for detecting fibronectin, vitronectin, α2, α3, β1 integrins, α-actinin, talin, and vinculin, or 5% SDS-PAGE for detecting laminin, αvβ3, and α5β1 integrins. The laminin B chains (B1 and B2) are present at 210 and 200 kD. A third B chain observed in a slightly lower molecular weight than B1 and B2 chains represents the laminin B subunit precursor forms (pB). Laminin A chain at 400 kD was not detected. (B) Flow cytometry analysis of β1 integrin subunit on cell surfaces. Y224 (solid line) or M634 (dotted line) cells cultured on plastic plate for 24 hours were incubated with the anti-β1 integrin antibody, and then a secondary antibody conjugated to fluorescein, and finally subjected to analysis by flow cytometry. Figure 5 Morphogenesis of various MDCK sublines cultured in 0.1% collagen gel. Morphology of MDCK sublines M634 (A), Y224 (B), M132 (C), M343 (D), M411 (E), and M532 (F) cells cultured in 0.1% collagen gels for eight days (scale bar 100 μm). Figure 6 Comparison of α2, α3, and αvβ3 integrin levels in various clones of MDCK cells. These integrins were assessed by Western blot analysis. Among these M clones, M634 cells exhibited the highest levels of either α2 or α3 integrins as well as tubulogenic capacity. The level of αvβ3 integrin does not seem to correlate with the potential of tubule morphogenesis. Figure 7 Expression of α3 integrin protein (A) and mRNA (B) in α3 integrin antisense RNA stably transfected M634 cells. (A) Western blot analysis of α3 and α2 integrin proteins in control and antisense transfectants. From the left: lane 1, pSG5α3(-)#2; lane 2, pSG5α3(-)#5; lane 3, pSG5α3(-)#7; lane 4, pSG5α3(+) control. The arrowheads show the position of the 215 and 105 kD protein standards. These lines contain an equal amount of α2 integrin, but their α3 integrin levels are differentially down-regulated, indicating that transfection has been successfully achieved. (B) Northern blot analysis to detect α3 integrin mRNA levels in pSG5α3(+) sense and pSG5α3(-) antisense constructs. The ribosomal (28s) RNA assessed by ethidium bromide staining was also shown as an internal control to ensure equal loading of total RNA. The expression of α3 integrin in these transfectants correlates well with their mRNA levels. Figure 8 Comparison of colony morphology (A-C), morphogenesis (D-F), and migration capacity (G) in α3 integrin antisense transfectants. Morphology of pSG5α3(+) control cells (A) and antisense expressing pSG5α3(-)#2 (B), and #7 cells (C) cultured on normal plastic plates is shown. Cell scattering is markedly reduced in pSG5α3(-)#2 cells, which exhibit the lowest levels of α3β1 integrin. (D-F) Morphogenesis of pSG5α3(+) control cells (D) and pSG5α3(-)#2 (E) and #7 cells (F) cultured in 0.1% collagen gel for eight days. The pSG5α3(-)#2 cells developed solely cell aggregates in 0.1% collagen gel, whereas pSG5α3(-)#7 cells exhibited shorter tubules with less branching than control cells. (A-F) Photographs were taken by phase-contrast microscopy. (G) Assessment of collagen-induced migration capacity in various pSG5α3(-) transfectants. Data are shown as percentage control migration capacity using pSG5α3(+) cells as the control. Each bar indicates mean ± SE of three experiments in triplicate (scale bar, 100 μm).
