FIGURE 1 Immunohistochemical staining for anti-Müllerian hormone (A, E), cytokeratin-18 (CK-18; B, C), and vimentin (D, F). In all cases, antibody binding was visualized using DAB chromogen as a substrate to generate a brown color. Sections were counterstained with Mayer’s hematoxylin, which stains nuclei blue. SC = Sertoli cell; SPG = spermatogonia; SPC = spermatocyte; RTD = round spermatid; SPZ = mature spermatids. (A, B), Focal spermatogenesis. Immunolabeling by anti-Müllerian hormone antibody (A) and by CK-18 antibody (B). The immunoreactivity (brown staining) is confined to Sertoli cell cytoplasm and is shown in the tubules in which spermatogenesis is arrested. The tubules demonstrating full spermatogenesis, in other words, the presence of mature spermatids, are negative for the expression of both markers. (C), Spermatocyte maturation arrest Immunolabeling by the CK-18 antibody. Sertoli cell cytoplasm is immunonegative. (D), Spermatocyte maturation arrest immunolabeling by the vimentin antibody. Consecutive section to that shown in (C), the left tubule. The Sertoli cell cytoplasm is uniformly immunoreactive. (E), Normal spermatogenesis. Immunolabeling by the anti-Müllerian hormone antibody. Sertoli cell cytoplasm is negative for anti-Müllerian hormone expression. (F), Normal spermatogenesis. Immunolabeling by the vimentin antibody. Consecutive section to that shown in (E). The Sertoli cell cytoplasm is uniformly immunoreactive. Original magnifications for A–C and E–F, ×400; original magnification for D, ×600. Bar = 50 μm. Maymon. Sertoli cells in spermatogenesis. FIGURE 2 Quantification of anti-Müllerian hormone (AMH) and cytokeratin-18 (CK-18) expression in testicular biopsies in the groups of azoospermic men revealing focal spermatogenesis (group A), spermatocyte maturation arrest (group B), or normal spermatogenesis (group C). Values are mean ± SEM. n = number of patients. Maymon. Sertoli cells in spermatogenesis. Fertil Steril 2002. Fertility and Sterility 2002 77, 904-909DOI: (10.1016/S0015-0282(02)03060-1)
