Along with expression of oncogenic KRAS, KLF4 expression in ADM leads to the development of premalignant PanIN lesions. However, KLF4 is downregulated in pancreatic ductal adenocarcinoma (PDA) and has tumor-suppressive properties, suggesting that premalignant changes and malignant progression in the pancreas are regulated by KLF4 through distinct targets and pathways.
