Figure 1 KLF4 Induces Cell Fate Changes and the Early Stages of Pancreatic Carcinogenesis KLF4 is upregulated in acinar cells during inflammation and/or injury and drives acinar cells toward a duct-like lineage (acinar-ductal metaplasia, ADM). Along with expression of oncogenic KRAS, KLF4 expression in ADM leads to the development of premalignant PanIN lesions. However, KLF4 is downregulated in pancreatic ductal adenocarcinoma (PDA) and has tumor-suppressive properties, suggesting that premalignant changes and malignant progression in the pancreas are regulated by KLF4 through distinct targets and pathways.
