Figure 1 The Igf2-Controlling KRAB-ZFP Duet in Murine ESCs In the wild-type cellular context, at the maternal allele, CTCF is recruited to the unmethylated Igf2/H19 ICR and blocks Igf2 promoter activation by distal enhancer. At the paternal allele, ZFP57 is recruited at the methylated Igf2/H19 ICR, allowing activation of Igf2 promoters. However, P0 is silenced and P1–P3 are dampened by ZFP568-induced repression. In ZFP568 KO embryonic stem cells (ESCs) at the paternal allele, the P0 promoter is unleashed and P1–P3 are further activated, resulting in increased IGF2 levels.
